Introduction
Hydrophilic Interaction Chromatography (HILIC) is a technique dedicated to the separation of polar and/or ionic compounds. Opposite to revered-phase (RP) mode separation, HILIC uses polar stationary phases and high
organic solvents comparable to normal phase (NP) mode. The polar analytes can partition into and out of the adsorbed water layer of the surface and can undergo ionic cation exchange interaction with silanol groups. In sum this will
lead to an enhanced retention of polar compounds and results in high loading capacities of those. While in RP-mode retention increases as ACN% decrease; in HILIC- mode the retention strongly increases as ACN% increase. Thus,
the elution of polar compounds show complementary selectivity in HILIC- compared to the separation in RP-mode. Fuji Silysia Chemical Ltd. (FSC) has developed two new media for HILIC mode named “HLC” Silica (Patent
applied). Both HLC Silicas have bipolar functional groups, covalent bonded on the silica surface. “HLC-A” can be applied for separation and large scale purification of a brought range of hydrophilic compounds while “HLC-
P” is dedicated to the separations of amino acids and peptides.

HLC-P: carboxyl chain with carbon amid bonding having a tertiary Amide.
HLC-A: di-carboxyl chain with two carbon amid bonding (tertiary and secondary Amides).
